Can KM be used on laptop and Desktop with one license?

Can KM be used on laptop and Desktop with one license? Or do I need to purchase two separate licenses?

Keyboard Maestro is licensed on a per user basis on up to five Macs. So one person can use it on a Mac and desktop with a single license.

Note that if you have a lab or similar, then you can alternatively license it as one license per Mac. But for most people, one license per user is the normal approach.

How can I set it up so I’m using the same macros I’ve created rather than having to recreate them on my laptop from scratch?

Follow those steps, creating a keyboardMaestroSync.kmsync file on your first computer, and store it in Dropbox (or similar), then sync the second computer. After that you can work and edit macros on either, and your changes will be synced.
